RESUMEN

Coffee (Coffea L.) is one of the main crops produced globally. Its contamination by the fungus Hemileia vastatrix Berkeley and Broome has been economically detrimental for producers. The objective of this work was to extract and characterize the essential oils from Eucalyptus citriodora Hook, Eucalyptus camaldulensis Dehn and Eucalyptus grandis Hill ex Maiden, produce and characterize nanoparticles containing these essential oils and evaluate the in vivo and in vitro antifungal activity of free and nanoencapsulated essential oils. The principal constituent of the essential oil from E. citriodora was citronellal; that from E. grandis was α-pinene; and that from E. camaldulensis was 1,8-cineol. The in vitro antifungal activity against the fungus H. vastatrix was 100% at a concentration of 1000 µl l-1 for all the oils and nanoparticles containing these natural products. The sizes of the nanoparticles produced with the essential oils from E. citriodora, E. camaldulensis and E. grandis were 402·13 nm, 275·33 nm and 328·5 nm, respectively, with surface charges of -11·8 mV, -9·24 mV and - 6·76 mV, respectively. Fourier transform infrared analyses proved that the encapsulation of essential oils occurred in the polymeric matrix of poly(ε-caprolactone). The incorporation of essential oils into biodegradable poly(ε-caprolactone) nanoparticles increased their efficiency as biofungicides in the fight against coffee rust, decreasing the severity of the disease by up to 90·75% after treatment with the nanoparticles containing the essential oil from E. grandis.

Essential oils encapsulated in a polymeric matrix can be used as an alternative method to control fungi and mycotoxins. The essential oils were extracted by hydrodistillation and characterized by gas chromatography. The nanofibres were produced from poly (acid lactic) (PLA) containing essential oils by the Solution Blow Spinning method. The antifungal and antimicotoxygenic properties were evaluated against Aspergillus ochraceus and Aspergillus westerdijkiae by the fumigation method. Terpinen-4-ol (20·23%), sabinene (20·18%), 1·8-cineole (16·69%) and γ-terpinene (11·03%) were the principal compounds present in the essential oil from Alpinia speciosa, whereas citral (97·67%) was dominant from Cymbopogon flexuosus. Microscopy images showed that the addition of essential oils caused an increase in the diameter of the nanofibres. The infrared spectroscopy results indicated the presence of essential oils in the PLA nanofibres. Differential scanning calorimetry curves also indicated the existence of interactions between the essential oils and polymeric macromolecules through their plasticizing action. The hydrophobic character of nanofibres was revealed by the contact angle technique. An antifungal effect was observed, the mycelial growths (3·25-100%) and the synthesis of ochratoxin A (25·94-100%) were inhibited by the presence of the nanofibres. The results suggest that bioactive nanofibres hold promise for application to control toxigenic fungi.

The extraction and characterization of the essential oils (EO) from Satureja montana L., Myristica fragrans H. and Cymbopogon flexuosus and the determination of their antibacterial and antioxidant activities were achieved. The EO were identified by gas chromatography/mass spectrometry and quantified by gas chromatography using a flame ionization detector. The antibacterial potential against Escherichia coli and Staphylococcus aureus was evaluated by cell susceptibility assays and by scanning electron microscopy. The antioxidant activity was evaluated by the 2,2-diphenyl-1-picrylhydrazyl assay, by ß-carotene bleaching and by determining the reducing power. Borneol (36·18%), γ-terpineol (12·66%) and carvacrol (11·07%) were the principal components in the EO from S. montana, and sabinene (49·23%) and α-pinene (13·81%) were found in the EO from M. fragrans. Geranial (59·66%) and neral (38·98%) isomers were the only major components in the EO from C. flexuosus. The EO from S. montana was effective against E. coli, with minimum inhibitory and bactericidal concentrations (MIC and MBC) of 6·25 µl ml-1 , whereas bactericidal potential against both was observed for the EO from M. fragrans; MIC = 6·25 µl ml-1 for S. aureus and MBC = 12·5 µl ml-1 for E. coli. A significant protective role on lipid substrates in the ß-carotene bleaching assay was seen for the EO from S. montana and M. fragrans. Overall, such EO can be promising agents against pathogenic bacteria and for protecting biomolecules during oxidative stress.

Natural enzyme inhibitors have been widely described in literature because of its pharmacological and cosmetic applications. Fungi found in caves represent a promising source of bioactive substances that are still little explored scientifically. Thus, the present work evaluated the presence of enzymatic modulators in a filtrate obtained from the cultivation of the cave fungus Lecanicillium aphanocladii (Family: Cordycipitaceae). Snake venoms from Bothrops alternatus and Bothrops atrox were used as an enzymatic source for the induction of the phospholipase, proteolytic, thrombolytic, cytotoxic and coagulant activities. Compounds present in the fungal filtrate inhibited 50, 23·8, 26·6, 50·9 and 52·5% of the proteolytic, phospholipase, haemolytic, thrombolytic and coagulant activities respectively. The filtrate was not cytotoxic on erythrocytes, but induced partial dissolution of thrombi. Fungal enzyme inhibitors that have low or no toxicity and can be obtained on a large scale and at low cost are relevant in the medical-scientific context. Therefore, the inhibition of phospholipases A2 and proteases observed in the present work highlights the potential of fungal metabolites for the development of drugs that can be used in the treatment of haemostasis and inflammation-related disorders. SIGNIFICANCE AND IMPACT OF THE STUDY: In this study, secondary metabolites synthesized by Lecanicillium aphanocladii, a fungus isolated from caves, demonstrated modulating action on proteases and phospholipases A2 present in snake venoms of the Bothrops genus, widely used as tools for the study of pathophysiology processes related to haemostasis and inflammation. The results suggest the possibility of future applications for these metabolites in the development of pharmaceuticals of medical-scientific interest.

BACKGROUND: Lower urinary tract dysfunction (LUTD) is a broad term describing the full spectrum of disorders in any of the stages of bladder function-storage or voiding LUTD is a clinical condition associated with emotional and behavioral disorders in children. This cross-sectional study aimed to investigate the association of emotional and behavioral symptoms and their impact on the quality of life (QoL) in children and adolescents with LUTD. METHODS: Eighty-eight patients and their parents enrolled in an interdisciplinary program for children and adolescents with LUTD were included in the analysis. Child Behavior Checklist (CBCL) was used to estimate the prevalence of behavioral and emotional problems through the assessment of 14 items. Pediatric Quality of Life Inventory (versions for parents and children) was applied in the versions for parents and children to evaluate the QoL. The Pediatric Quality of Life Inventory-PedsQL, version 4.0-was applied separately for parents and patients. The association of clinical variables and aspects related to QoL of patients were evaluated through non-parametric correlations (Spearman) and multiple linear regression analysis. RESULTS: According to CBCL's clinical scores, 56% of the patients showed total behavioral problems, 55% internalizing, and 38% externalizing. When comparing the conditions of LUTD and the CBCL scores, patients with voiding postponement had the lowest rates of total problems (P = 0.036). Children and adolescents with LUTD who also had enuresis showed a higher frequency of externalizing problems (P = 0.001), especially aggressive behavior (P = 0.013). Scores of patients with LUTD were significantly lower in all domains of QoL than normative data. Presence of behavioral problems was associated with worse QoL in all evaluated aspects. The total QoL was most influenced by the CBCL school competence scale according to the regression model analysis. CONCLUSIONS: The study findings suggest the relevance of evaluation of behavioral and social repercussions of LUTD to improve the multidisciplinary approach for this condition in pediatric population.

Os óleos essenciais são metabólitos secundários dos vegetais com propriedades biológicas diferenciadas, dentre elas, a atividade contra microrganismos, sendo de interesse da indústria de alimentos as concentrações mínimas inibitórias (CMI) desses óleos para diversas bactérias. As CMI variam em função dos compostos majoritários e da espécie de bactéria. Nesta pesquisa, os óleos essenciais das plantas Satureja montana L., Cymbopogon nardus L. e Citrus limonia Osbeck foram caracterizados quimicamente e se determinou a CMI sobre as bactérias Staphylococcus aureus ATCC 2592 e Escherichia coli ATCC 25922. A CMI dos óleos testados contra E. coli e S. aureus foi de 1,5%, exceto para o óleo essencial de S. montana sobre S. aureus, a qual foi sensível a este óleo a partir da concentração de 5,0%. Sobre a constituição química, os componentes majoritários dos óleos de S. montana, C. narduse C. limonia Osbeck foram respectivamente o timol, citronelal e limoneno.


Essential oils are plant secondary metabolites with different biological properties, such as activity against microorganisms, being of major interest to the food industry. The minimal inhibitory concentrations (MIC) of these oils for various bacteria should be determined. The MICvaries according to the oil used by the major compounds and phenotypic and genotypic characteristics of the bacteria. In this research, the essential oils of the plants Satureja montana L., Cymbopogon nardus L. and Citrus limonia Osbeck were chemically characterized and the MIC of the bacteria Staphylococcus aureusATCC 2592 and Escherichia coli ATCC 25922, determined. The MICfor all oils against E. coli was 1.5%, and S. aureus was sensitive to the concentration of 5.0% of S. montanaoil and tothe concentration of 1.5% of the essential oils of the other plants.Chemically, the major components ofthe S. montana, C. nardus and C.limonia Osbeck oils were respectively thymol, citronellal and limonene.

This study sought to evaluate the chemical composition of the Allium sativum and Origanum vulgare essential oils and their effect on the growth inhibition of microorganisms, such as P. aeruginosa, S. Choleraesuis, A. flavus, A. niger and P. simplicissimum, important food contaminants. The main constituents of the oregano essential oil were 4-terpineol (27.03%), γ-terpinene (20.04%), and β-cymene (6.34%), and the main constituents of the garlic essential oil were diallyl trisulfide (38, 81%), diallyl disulfide (25.23%), and methyl allyl trisulfide (12.52%). Inhibition zones were formed in in vitro tests on the bacteria S. Choleraesuis and P. aeruginosa, except for A. sativum against P. aeruginosa. The inhibition of mycelial growth caused by the oregano essential oil occurred with the concentrations of 0.10, 0.03 and 0.05 mg mL-1 for the A. flavus, A. niger and P. simplicissimum fungi, respectively. The CMI for the garlic oil began at the 0.03 mg mL-1 concentration for all species of fungi. The oils presented an inhibitory effect against the microorganisms studied and constitute an alternative for microbiological control in food.


Objetivou-se avaliar a composição química e o efeito inibitório dos óleos essenciais de Allium sativum e Origanum vulgare frente ao crescimento dos micro-organismos Pseudomonas aeruginosa, Salmonella Choleraesuis, Aspergillus flavus, Aspergillus niger e Penicillium simplicissimum, importantes patógenos causadores de contaminações em alimentos. Para quantificação e identificação dos constituintes químicos dos óleos, utilizou-se cromatógrafo gasoso acoplado a espectrômetro de massas. Os principais constituintes do óleo essencial de orégano foram o 4-terpineol (27,03%), γ-terpineno (20,04%), β-cimeno (6,34%), e do alho, o dialil trissulfeto (38,81%), dialil dissulfeto (25,23%), metil alil trissulfeto (12,52%). Os resultados dos testes in vitro sobre as bactérias S. Choleraesuis e P. aeruginosa indicaram a formação de halo de inibição e revelaram o efeito inibitório para os referidos óleos, exceto para o óleo de A. sativum frente a P. aeruginosa. Para os fungos A. flavus, A. niger e P. simplicissimum a inibição do crescimento micelial provocada pelo óleo essencial de orégano ocorreu a partir das concentrações de 0,10, 0,03 e 0,05 µg mL-1, respectivamente, sendo que a CMI para o óleo de alho iniciou-se a partir da concentração 0,03 µg mL-1 para todas as espécies de fungos. Foi possível verificar que os óleos possuem efeito inibitório sobre os microrganismos estudados, sendo, portanto, uma alternativa no controle microbiológico de alimentos.

A morfofisiologia relacionada à absorção de ácidos graxos voláteis (AGV) ao longo do trato gastrintestinal de ruminantes não é totalmente caracterizada. Desse modo, os objetivos deste trabalho foram mensurar a extensão da superfície de absorção e determinar o índice mitótico (IM) do abomaso, do ceco e da alça proximal do cólon ascendente (APCA). Dez bovinos mestiços adultos tiveram seu estômago e intestino grosso removidos imediatamente após o abate. A área total da superfície de absorção foi mensurada por meio de digitalização e análise de imagens. Cortes histológicos foram feitos para determinação do IM. A superfície absortiva do abomaso, 0,58m², foi menor (P<0,01) do que a do rúmen, 6,53m², e a do omaso, 2,31m². A superfície absortiva do ceco e da APCA, 0,23m², correspondeu a 3,5% da superfície do rúmen e a 10% da superfície do omaso. O IM observado foi 0,48%; 0,14%; 0,36% e 0,41% para as regiões de pregas espirais, pilórica, ceco e APCA, respectivamente. Observou-se correlação positiva entre a massa tecidual do abomaso e a área de superfície de absorção, aspecto também observado no ceco-APCA. Foi possível estabelecer regressões para facilitar a mensuração da superfície absortiva do abomaso e do ceco-APCA.


The morphology and physiology related to volatile fatty acid (VFA) absorption throughout the gastrointestinal tract of ruminants is not totally characterized. The purpose of this work was to measure the abomasum and cecum-colon absorptive surface extension and determine the mitotic index (MI). Ten adult crossbred bovine had their stomach and large intestine removed after slaughter. The total area of the absorptive surface was measured through image capture and analysis. Histological sections were performed to measure the MI. The abomasum absorptive surface (0.58m²) was lower (P<0.01) than that of the rumen (6.53m²) and omasum (2.31m²). The cecum-colon absorptive surface (0.23m²) corresponded to 3.5% of the rumen and 10% of the omasum. The MI observed was 0.48%; 0.14%; 0.36% and 0.41% for the regions of spiral folds, pyloric, cecum and proximal loop of ascending colon, respectively. A positive correlation between abomasum tissue mass and the abomasum absorptive surface was observed. This phenomenon has also occurred in the cecum and colon. It was possible to establish regressions to facilitate measurements of the absorptive surface of the abomasum and cecum-colon.

As concentrações mínimas inibitórias (CMI) desses óleos para diversas bactérias devem ser determinadas. As CMI variam de acordo com o óleo utilizado, dos compostos majoritários e do tipo de bactéria. Nesta pesquisa, os óleos essenciais das plantas Satureja montana L., Cymbopogon nardus L. e Citrus limonia Osbeck foram caracterizados quimicamente e determinada a CMI sobre as bactérias Staphylococcus aureus ATCC 2592 e Escherichia coli ATCC 25922. A CMI para todos os óleos contra E. coli foi 1,5%, já S. aureus foi sensível a partir da concentração de 5,0% do óleo essencial de S. montana e 1,5% foi a CMI obtida quando utilizados os óleos essenciais das outras espécies estudadas. Na constituição química os componentes majoritários para os óleos de S. montana, C. narduse C. limonia Osbeck foram respectivamente o timol, citronelal e limoneno.

Meat quality traits were compared in Texel x Polwarth and Texel x Corriedale ram lambs, with live weight of 25kg, finished on pastures rich in white clover. The two crossbred groups showed similar results (P>0.05) for pH, moisture, protein, meat color, individual SFA and total SFA. Fat content was higher and cholesterol lower in Texel x Polwarth lambs (P<0.05), which also had higher amounts of MUFA (P<0.05) and lower levels of total PUFA and n-6 PUFA (P<0.01). Differences in meat quality among the two crossbred groups were minor, and possibly not perceptible from the consumer standpoint. However, both groups of lambs produced lean meat with high amounts of PUFA (>16%), possibly due to the ingestion of white clover in the finishing period. Overall, these results indicate that a differentiated product can be obtained in these conditions, with higher nutritional value than conventional lamb meat.

Objetivou-se identificar e quantificar os constituintes e avaliar a atividade antimicrobiana dos óleos essenciais de Mentha piperita, Cymbopogon citratus, Ocimum basilicum e Origanum majorana contra cepas de Escherichia coli enteropatogênica, Salmonella enterica Enteritidis, Listeria monocytogenes e Enterobacter sakazaki. A obtenção dos óleos essenciais foi realizada a partir de folhas secas, empregando-se a técnica de hidrodestilação e utilizando-se a aparelho de Clevenger modificado. A atividade antibacteriana dos óleos essenciais foi determinada pelo método de difusão em ágar. Observou-se que os óleos essenciais inibiram o crescimento bacteriano, mas a efetividade foi variada. Entre os óleos essenciais testados, M. piperita apresentou maior atividade antibacteriana para E. coli, (8.106 UA mL-1) quando comparada as demais bactérias, atividade moderada para Salmonella enterica Enteritidis e Enterobacter sakazakii (1.706 e 3.200 UA mL-1 respectivamente) e baixa atividade para Listeria monocytogenes (106,67 UA mL-1). Já óleo essencial de Cymbopogon citratus apresentou maior atividade antimicrobiana frente a E. coli (9.386 UA mL-1) e atividade moderada frente a Enterobacter sakazakii, Salmonella enterica Enteritidis e Listeria monocytogenes (2.773 UA mL-1 para ambas). Ocimum basilicum apresentou maior atividade antibacteriana frente E. coli e Enterobacter sakazakii (6.826 e 8.106 UA mL-1 respectivamente), moderada atividade frente a Salmonella enterica Enteritidis (1.600 UA mL-1) e não apresentou atividade frente a Listeria monocytogenes.Origanum majorana também foi testado neste estudo e apresentou maior atividade antimicrobiana frente E. coli (5.973 UA mL-1), atividade moderada para Salmonella enterica Enteritidis e Enterobacter sakazakii (1.706 e 2.346 UA mL-1 , respectivamente) e não apresentou atividade para Listeria monocytogenes.


ABSTRACT The objective of this work was to identify and quantify the constituents, and to evaluate the antimicrobial activity of the essential oils from Mentha piperita, Cymbopogon citratus, Ocimum basilicum and Origanum majorana, against enteropathogenic Escherichia coli, Salmonella enterica Enteritidis, Listeria monocytogenes and Enterobacter sakazakii. The essential oils were obtained from dried leaves by using the hydrodistillation technique and the modified Clevenger apparatus, and their bacterial activity was determined by using the agar diffusion technique. The essential oils inhibited bacterial growth, but their effectiveness was varied. Among the essential oils tested, that from M. piperita showed a greater antimicrobial activity against E. coli (8.106 UA mL-1), moderate activity for S. enterica Enteritidis and E. sakazakii (1.706 e 3.200 UA mL-1 respectively) and low activity for L. monocytogenes (106,67 UA mL-1). However, the essential oil from C. citratus presented a greater antimicrobial activity against E. coli (9.386 UA mL-1) and a moderate activity against E. sakazakii, S. enterica Enteritidis and L. monocytogenes (2.773 UA mL-1 for both). The essential oil from O. basilicum showed a greater antimicrobial activity against E. coli and E. sakazakii (6.826 e 8.106 UA mL-1 respectively),moderate activity against S. enterica Enteritidis (1.600 UA mL-1), and was inactive against L, monocytogenes. Origanum majorana, which was also tested in our work, showed a greater antibacterial activity against E. coli, (5.973 UA mL-1) moderate activity against S. enterica Enteritidis and E. sakazakii (1.706 e 2.346 UA mL-1 , respectively), and was inactive against L. monocytogenes.

O presente trabalho teve como objetivo a caracterização de plantas frescas e secas (comerciais) de alfavaca, orégano e tomilho, a obtenção dos óleos essenciais através do método de arraste a vapor e a quantificação dos compostos químicos por CG/EM. As plantas frescas e as secas comerciais foram submetidas às análises de umidade, extrato etéreo, proteína, fibra bruta, cinzas, extrato não nitrogenado, valor calórico, teor de óleo essencial e identificação dos compostos majoritários através da cromatografia gasosa-espectrometria de massas. Dentre a caracterização obtida os resultados na base seca mostraram-se promissores, sendo o teor de proteína e de cinzas na alfavaca seca comercial com 17,34 g 100 g-1 e 8,12 g 100 g-1, respectivamente; a fibra bruta no orégano seco comercial com 15,65 g 100 g-1; o extrato etéreo, o extrato não nitrogenado e o valor calórico no tomilho seco comercial com 9,30 g 100 g-1, 52,72 g 100 g-1 e 356,74 Kcal 100 g-1, respectivamente. Obteve-se o maior rendimento de óleo essencial na alfavaca seca comercial com 1,02%, enquanto a alfavaca fresca apresentou o menor rendimento, com apenas 0,13%. Na alfavaca fresca encontrou-se 87,38% de eugenol e 6,27% de timol, enquanto na alfavaca seca comercial observou-se redução no eugenol (71,12%) e aumento do timol (13,28%). No orégano fresco foram quantificados quatro picos o γ-terpineno (33,45%), 4-terpineol (25,59%), timol (14,21%) e carvacrol (2,30%). Já no óleo essencial de orégano seco comercial houve redução no γ-terpineno (28,73%) e aumento no 4-terpineol (27,58%), timol (19,71%) e carvacrol (3,67%). No óleo essencial do tomilho fresco foram quantificados três picos o borneol (66,66%), timol (13,41%) e linalol (3,24%). Por outro lado, no óleo essencial do tomilho seco comercial houve redução no borneol (37,90%) e aumento no timol (20,61%) e linalol (10,34%). Pode-se concluir que as folhas secas comerciais analisadas de alfavaca, orégano, e tomilho apresentam potencial para o enriquecimento dos alimentos ou para a obtenção dos óleos essenciais.


This study aimed to characterize commercial fresh and dry medicinal plants (basil, oregano and thyme), to obtain essential oil by the steam distillation method and to quantify chemical compounds by means of GC/MS. The fresh and dry plants were subjected to the following analyses moisture, ether extract, protein, crude fiber, ash, non-nitrogenous extract, caloric value, essential oil content and identification of major compounds by gas chromatography-mass spectrometry. Considering the obtained characterization, the following results on dry basis proved promising: protein and ash content in commercial dry basil with 17.34 g 100 g-1 and 8.12 g 100 g-1, respectively; crude fiber in commercial dry oregano with 15.65 g 100 g-1; ether extract, non-nitrogenous extract and caloric value in commercial dry thyme with 9.30 g 100 g-1, 52.72 g 100 g-1 and 356.74 Kcal 100 g-1, respectively. The highest essential oil yield was obtained for commercial dry basil with 1.02% and the lowest yield was obtained for fresh basil with only 0.13%. Chromatography indicated 87.38% eugenol and 6.27% thymol in fresh basil. For commercial dry basil, the chromatogram showed a reduction in eugenol (71.12%) and an increase in thymol (13.28%). Four peaks were quantified for fresh oregano the γ-terpinene (33.45%), 4-terpineol (25.59%), thymol (14.21%) and carvacrol (2.30%). For the essential oil of commercial dry oregano, there was a decrease in γ-terpinene (28.73%) and an increase in 4-terpineol (27.58%), thymol (19.71%) and carvacrol (3.67%). In the chromatogram of the essential oil of fresh thyme, three peaks were quantified: borneol (66.66%), thymol (13.41%) and linalool (3.24%). On the other hand, in the chromatogram of the essential oil of commercial dry thyme, there was a decrease in borneol (37.90%) and an increase in thymol (20.61%) and linalool (10.34%). It can be concluded that commercial dry leaves of basil, oregano and thyme are feasible to enrich foods or to obtain essential oils.

Objetivou-se determinar o rendimento, a composição química e avaliar a atividade antilisterial dos óleos essenciais de Cymbopogon citratus (DC.) Stapf. (capim-limão) e Cymbopogon nardus (L.) Rendle (capim-citronela), puros e em combinação. A obtenção dos óleos essenciais foi realizada a partir de folhas frescas, empregando-se a técnica de hidrodestilação e utilizando-se aparelho de Clevenger modificado. Os rendimentos foram calculados a partir da massa obtida na extração e da umidade do material vegetal. A análise qualitativa foi realizada por cromatografia em fase gasosa acoplada à espectrometria de massas e a avaliação dos teores dos constituintes químicos por cromatografia em fase gasosa com detector de ionização de chamas. O efeito bacteriostático de diferentes concentrações dos óleos essenciais, puros e em combinação, foi determinado pela técnica de difusão em ágar. As concentrações mínimas inibitórias (CMIs) foram utilizadas na determinação do tempo de ação bactericida. O rendimento, em base seca, foi de 1,39 por cento (v/p) para o óleo essencial de C. citratus e de 2,27 por cento (v/p) para o óleo essencial de C. nardus. Geranial e neral foram os constituintes majoritários do óleo essencial de C. citratus, enquanto, para C. nardus, foram citronelal, geraniol e citronelol. A maioria das concentrações utilizadas mostrou-se efetiva contra Listeria monocytogenes e observaram-se efeitos bactericidas nas CMIs. Óleos essenciais de C. citratus e C. nardus, puros e em combinação, constituem novas alternativas de antibacterianos naturais a serem utilizados no controle de L. monocytogenes na indústria de alimentos.


The aim of this study was to determine the yield and the chemical composition of essential oils from Cymbopogon citratus (DC.) Stapf. (lemongrass) and Cymbopogon nardus (L.) Rendle (citronella), and to evaluate their antilisterial activity when pure and combined. The essential oils were obtained from fresh leaves through hydrodistillation by using a modified Clevenger-type device. Yield was calculated based on the extracted mass and the plant humidity. The qualitative analysis was done by gas chromatography-mass spectrometry and the levels of the chemical components were detected by gas chromatography with flame ionization detector. The bacteriostatic effect of different concentrations of pure and combined essential oils was determined by the agar diffusion technique. Minimum inhibitory concentrations (MICs) were used to determine the time of bactericidal action. The yield was 1.39 percent (v w-1 dry basis) for C. citratus essential oil and 2.27 percent (v w-1) for C. nardus essential oil. Geranial and neral were the major components of C. citratus essential oil, while citronellal, geraniol and citronellol were predominant in C. nardus oil. Most of the used concentrations were efficient against Listeria monocytogenes and bactericidal effects were observed for the MICs. Essential oils of C. citratus and C. nardus, pure and combined, constitute new alternatives of natural antibacterials for the control of L. monocytogenes in the food industry.

Essential oils of Origanum majorana L. (marjoram), Illicium verum Hook. f. (star-anise) and Cinnamomum zeylanicum Blume (cinnamon) were obtained by steam distillation using a modified Clevenger device. The antimicrobial activity of each oil was evaluated against the bacteria Staphylococcus aureus, Escherichia coli and the fungi Aspergillus flavus and Aspergillus parasiticus by observing their growth and/or mycelial inhibition through comparison with the standard dish (without oil). The essential oils were analyzed using a gas chromatograph coupled to a mass spectrometer for identification and coupled to a flame ionization detector for quantification. The major constituents of marjoram, star-anise and cinnamon essential oils were 4-terpineol, trans-anetole and cinnamic aldehyde, respectively. In in vitro tests, essential oils of marjoram and cinnamon promoted an inhibitory effect on the bacteria S. aureus and E. coli, while the essential oil of star-anise presented activity only against E. coli. Marjoram, star-anise and cinnamon oils were effective against the studied fungi, presenting an inhibitory effect. The minimal inhibitory concentration for the mycelial growth of A. parasiticus was 1 and 0.01 µL mL-1 for star-anise and cinnamon oils, respectively. The minimal inhibitory concentration for A. parasiticus was 0.25, 2 and 2 µL mL-1 for cinnamon, star-anise and marjoram oils, respectively.


Óleos essenciais de Origanum majorana L. (manjerona), Illicium verum Hook. f. (anis estrelado) e Cinnamomum zeylanicum Blume (canela) foram obtidos pela técnica de arraste a vapor d'água com aparelho de Clevenger modificado. Foram avaliadas as atividades antimicrobianas de cada um sobre as bactérias Staphylococcus aureus, Escherichia coli e para os fungos Aspergillus flavus e Aspergillus parasiticus, observando o crescimento e/ou inibição micelial, comparando-se estes com a placa-padrão (sem óleo). Os óleos essenciais foram analisados em cromatógrafo gasoso acoplado a espectrômetro de massa para a identificação e cromatógrafo gasoso com detector de ionização de chamas para a quantificação dos compostos. Os principais constituintes dos óleos essenciais de manjerona, anis-estrelado e canela foram o 4-terpineol, trans-anetol e aldeído cinâmico, respectivamente. Nos testes in vitro, os óleos essenciais de manjerona e canela promoveram efeito inibitório sobre as bactérias S. aureus e E. coli, enquanto o óleo essencial de anis estrelado apresentou atividade apenas frente E. coli. Os óleos de manjerona, anis estrelado e canela foram efetivos sobre os fungos estudados, apresentando efeito inibitório. A concentração mínima inibitória pra o crescimento micelial de A. parasiticus foi de 1 e 0,01 µL mL-1 para os óleos de anis-estrelado e canela, respectivamente. Enquanto a concentração mínima inibitória para A. parasiticus foi de 0,25; 2 e 2 µL mL-1 para os óleos de canela, anis-estrelado e manjerona, respectivamente.

Este trabalho teve como objetivos avaliar a composição química do óleo essencial de Baccharis tridentata Vahl, as atividades antioxidante e fungitóxica, e estudar a morfologia das estruturas secretoras do óleo essencial presentes na superfície foliar por meio de microscopia eletrônica de varredura (MEV). A extração do óleo essencial foi realizada por hidrodestilação, as análises quantitativas e qualitativas foram executadas por meio de cromatografia em fase gasosa com detector de ionização de chamas (FID) e acoplada à espectrometria de massas, respectivamente. A atividade antioxidante foi realizada empregando-se os métodos de redução do radical estável DPPH e o ensaio de oxidação do sistema β-caroteno/ácido linoleico. As atividades fungitóxicas foram avaliadas utilizando o teste bioanalítico in vitro, sobre a inibição do crescimento micelial dos fitopatógenos Fusarium oxysporum, Colletotrichum gloeosporioides e Rhizoctonia solani. A composição química revelou a presença de 28 compostos, sendo o α-tujeno (22,93 por cento) o constituinte majoritário; não foi observada atividade antioxidante por meio dos ensaios utilizados, no entanto, observou-se atividade fungitóxica sobre o crescimento micelial dos fitopatógenos estudados. Já os estudos da superfície foliar por MEV revelaram a presença de tricomas glandulares em ambas as superfícies abaxial e adaxial.


This study aimed to evaluate the chemical composition and the antioxidant and fungitoxic activities of Baccharis tridentata essential oil, as well as to study the morphology of its secretory structures present on the leaf surface by scanning electron microscopy (SEM). The essential oil was extracted by hydrodistillation; the quantitative and qualitative analyses were performed on a gas chromatograph equipped with a flame ionization detector (FID) and coupled to a mass spectrometer, respectively. The antioxidant activity was determined by the methods of reduction of the DPPH stable radical and oxidation of the β-carotene/linoleic acid system. Fungitoxic activities were assessed by the in vitro bioanalytical test on the inhibition of the mycelial growth of the plant pathogens Fusarium oxysporum, Colletotrichum gloeosporioides and Rhizoctonia solani. The chemical composition revealed the presence of 28 compounds, with α-thujene (22.93 percent) as the major constituent. No antioxidant activity was observed in the tests used; however, there was fungitoxic activity against the mycelial growth of plant pathogens. Leaf surface studies by SEM revealed the presence of glandular trichomes on both abaxial and adaxial surfaces.

Estudou-se a rentabilidade da atividade leiteira de 17 propriedades da região de Lavras/MG e identificaram-se os componentes que exerceram maior influência sobre os custos finais da atividade. Os dados foram coletados mensalmente durante o período de janeiro de 2004 a dezembro de 2005. Considerou-se a margem bruta, a margem líquida e o resultado (lucro ou prejuízo) como indicadores de eficiência econômica. Os itens componentes do custo operacional efetivo que exerceram maior influência sobre os custos da atividade leiteira foram, em ordem decrescente, alimentação (55,9 por cento), mão-de-obra (19,1 por cento), despesas diversas (11,3 por cento), energia (7,1 por cento), sanidade (4,9 por cento), ordenha (0,74 por cento), impostos (0,74 por cento) e inseminação artificial (0,23 por cento). A margem líquida e o resultado positivo indicam que os pecuaristas têm condições de produzir a médio e a longo prazo, com conseqüente capitalização. Os resultados do estudo não podem ser extrapolados por não se tratar de uma amostra probabilística.


It was studied the profitability of the milk production in 17 properties in the region of Lavras (MG), and the components that had the greatest influence on the final costs of the business were identified. The data were monthly collected from January 2004 to December 2005. The gross margin, net margin, and the result (profit or loss) were taken into consideration as indicators of economic efficiency. The component items of the effective operational cost that had the greatest influence upon the costs of dairy business were, in decreasing order, feeding (55.9 percent), labor (19.1 percent), different expenses (11.3 percent), energy (7.1 percent), health (4.9 percent), milking (0.74 percent), taxes (0.74 percent), and artificial insemination (0,23 percent). The net margin and the positive result show the farmers have conditions to produce in middle and long terms, with consequent capitalization. Although, the results of the study can not be extrapolated, since they do not represent a random sampling.

This study analyses the anti-proliferative effect of lemongrass essential oil and its main constituent (citral) on all 3 evolutive forms of Trypanosoma cruzi. Steam distillation was used to obtain lemongrass essential oil, with chemical composition determined by gas chromatography (GC) and GC coupled to mass spectrometry (GC-MS). The IC50/24 h (concentration that reduced the parasite population by 50%) of the oil and of citral upon T. cruzi was determined by cell counting in a Neubauer chamber, while morphological alterations were visualized by scanning and transmission electron microscopy. Treatment with the essential oil resulted in epimastigote growth inhibition with IC50=126.5 microg/ml, while the IC50 for trypomastigote lysis was 15.5 microg/ml. The IC50/48 h for the Association Index (% macrophage infection x number of amastigotes per cell) was 5.1 microg/ml, with a strong inhibition of intracellular amastigote proliferation. Ultrastructural analysis demonstrated cytoplasmic and nuclear extraction, while the plasma membrane remained morphologically preserved. Our data show that lemongrass essential oil is effective against T. cruzi trypomastigotes and amastigotes, and that its main component, citral, is responsible for the trypanocidal activity. These results indicate that essential oils can be promising anti-parasitic agents, opening perspectives to the discovery of more effective drugs of vegetal origin for treatment of parasitic diseases. However, additional cytotoxicity experiments on different cell lines and tests in a T. cruzi-mouse model are needed to support these data.

Acylsugars present in Lycopersicon pennellii are responsible for the high levels of pest resistance often found in this wild tomato taxon. We investigated the inheritance of acylsugar contents in segregating populations of the interspecific tomato cross L. esculentum x L. pennellii and estimated correlations between leaflet acylsugar contents and the levels of mite repellence. Acylsugar contents were quantified with the Sommogy-Nelson colorimetric method in the acessions L. esculentum 'TOM-584' (P(1), low acylsugars), L. pennellii 'LA-716' (P(2), high acylsugars), in the interspecific F(1) (P(1) x P(2)) and in the F(2 )(P(1) x P(2)) generations. Mite resistance was assessed by a repellence test. Broad-sense heritability of acylsugar contents was moderately high (h(2)(b) = 0.476). Frequency distributions in the P(1), P(2), F(1) and F(2) can be explained by the action of a single major locus, with near-complete dominance of the L. esculentum allele for low-acylsugar content over the L. pennellii allele for high content. Indirect selection for high levels of acylsugars in leaflets led to correlated increases in the levels of mite repellency, indicating that acylsugars may be the main factor involved in mite resistance

This work evaluated in a population of heroin and heroin plus cocaine human addicts: 1. Norepinephrine (NE), epinephrine (Epi) and 3-methoxy-4-hydroxyphenylglycol (MHPG) (the principal metabolite of brain NE) plasma levels; 2. Monoamine oxidase (MAO) activity; and 3. 3H-imipramine specific binding to the amine carrier in platelets. NE plasma levels were significantly lower in the short-term heroin user groups (1-3 and 4-6 yr), a finding not observed in both long-term heroin user ( > 6 yr) and heroin plus cocaine user ( > 6 yr) groups. Epi levels changed in a similar manner, except that a significant increase was noted in heroin plus cocaine abusers. Conversely, dopamine and MHPG plasma levels increased with the duration of heroin use, and even more with cocaine abuse. Platelet MAO activity increased in all groups. Specific 3H-imipramine binding sites showed an increase after 3 yr of heroin abuse and in all heroin plus cocaine addicts. In conclusion, short-term use of heroin decreases NE or Epi release, but with prolonged use, a slow adaptation occurs. In contrast, cocaine inhibits the neuronal Epi uptake, even in a situation of long duration of abuse. Probably the amine levels additionally regulate the amine carrier, resulting in changes that show a different pattern from major depression. These drugs of abuse may also influence directly or indirectly related enzymatic systems.
